Quantitative study of fluorescent probes in cellular systems by flow cytometry.

ABSTRACT

We studied the in situ binding of Propidium Iodide (PI) to DNA and of Fluorescein Isothiocyanate (FITC) to proteins using flow cytometry. Experimental data were interpreted by means of mathematical models specific for the two staining reactions, and the binding parameters were obtained by best-fitting of the data. For the PI-DNA interaction, a model based on two classes of highly fluorescent sites with different affinity constants gave the most satisfactory fitting. The FITC-protein binding was adequately described by a single affinity constant. In both cases the number of binding sites turned out to be reduced with respect to non-in situ stainings reported in the literature. The present analysis allowed also to recognize staining conditions that do not assure linearity between fluorescence and substrate amount in flow cytometric measurements.
