Cell-mediated antigen presentation to Th-cells: a mathematical model.

ABSTRACT

In the framework of a model for the interaction between antibodies and conformation-dependent epitopes originating from the association of protomers into a multimeric antigen, we have examined how, on the surface of the antigen-presenting cells, the complex between the antigenic peptide and the major histocompatibility class II molecule intercacts with the T-cell receptor. Following the description proposed by Bell et al. of the thermodynamics of cell-to-cell adhesion (Bell et al., Biophys. J., 45, 1051, 1984), it is possible to determine whether the surface density of the interacting molecules and the association constant values are compatible with intercellular adhesion - a necessary prerequisite for T-cell activation. A relatively simple mathematical description of a rather complex phenomenon can thus be achieved.
